海岸林・山林復旧を目的とした耐塩性および強増殖性ショウロ菌の開発

Outline of Annual Research Achievements

本年度は,突然変異誘発法および土壌基質における増殖性評価法の確立を目指した.
突然変異誘発剤として広く用いられている ethylmethanesulfonate (EMS) がショウロ菌の突然変異誘発に有効であること,また, 20 mg/mLの濃度で処理することが適正であることを明らかにした.EMS処理ショウロ担子胞子から再生した一次菌糸体において,NaCl含有寒天培地で旺盛に成長した好塩性一次菌株5菌株を分離した.さらに好塩性の一次菌株同士を交雑することで交雑F1を育成し,塩ストレス耐性を評価した結果,4菌株の好塩性二次菌株,4菌株の耐塩性二次菌株を選抜できた.さらに,2菌株の塩感受性二次菌株も選抜できた.
次に,様々な土壌を用いてショウロ菌株の増殖状況を可視化できる基質を探索した.その結果,砂土壌基質を用いることでショウロ菌の増殖状況を明瞭に判定できることが判明した.次に,寒天培地と砂土壌基質における菌糸体の増殖速度を比較した結果,寒天培地で好塩性や耐塩性を示した菌株は人工海水を含む砂土壌基質で旺盛に増殖し,寒天培地で塩感受性を示した菌株は海水を含む砂基質では増殖しなかった.育成菌株を砂土壌基質に接種して増殖性と耐塩性について調査した結果,強増殖性および耐塩性の両特性を具備した菌株を2菌株選抜できた.
現在は,これらの菌株を人工的に宿主クロマツ実生根に接種し,共生したショウロ菌糸体の塩ストレスに対する反応性について解析中である.

Reason

ショウロ菌株における突然変異誘発法および土壌基質における増殖性評価法を確立できた.
一方,土壌基質で,5~10菌株の強増殖性菌株の作出を目指していたが,2菌株しか作出できなかった.しかし,砂土壌基質で増殖性と耐塩性を同時に評価できたことから,本2菌株は強増殖性および耐塩性の両特性を具備した有望菌株であるといえる.

Strategy for Future Research Activity

今後は,作出した強増殖性・耐塩性菌株および対照として塩感受性菌株を宿主クロマツ実生根に人工接種して感染苗木を育成し,感染苗木の塩ストレス耐性を評価することで,作出ショウロ菌株の宿主樹木への塩ストレス耐性付与効果を評価する.さらには,耐塩性を付与したショウロ感染苗木を東北地方の津波被災地の海岸林への導入を試みる.
