Outline of Annual Research Achievements

前年度までに、CCK8Sは脊髄内に分布するCCK-2R発現細胞を介してアロネーシスを発症することを見出した。本年度はin situ hybridization法により、C57BL/6JマウスのDRG及び脊髄におけるCCK mRNA及びCCK2R mRNAの発現分布について検討した。DRGにおいて、CCK mRNAは小型及び中型のDRG細胞において発現が確認された。また、脊髄においてもCCK mRNAの発現が検出され、これらCCK mRNA発現細胞はニューロンのマーカー遺伝子NeuNを発現していた。さらに、脊髄後角に発現するCCK受容体は主にCCK2Rであり、CCK1Rの発現レベルは非常に低いこと、CCK2R発現細胞は前述したNeuN mRNAを発現していることが明らかとなった。以上の結果から、CCK8Sの産生細胞はDRGニューロンあるいは脊髄後角ニューロンであり、これらのニューロンから遊離されたCCK8Sは、脊髄内のCCK2R発現ニューロンを介してアロネーシスの誘発に関与していることが示唆された。
