Outline of Annual Research Achievements |
今年度は、新たに2名の急性GVHD患者から皮膚等のGVHD生検組織と同時期の末梢血の提供を受けた。それら試料からRNAを抽出し、T細胞受容体(TCR)遺伝子塩基配列のdeep sequenceを行った。そして組織浸潤T細胞のTCR塩基配列と末梢血中T細胞のTCR塩基配列を照合、比較した。 本研究期間全体の研究実績をまとめる。計8名の急性GVHD患者から生検組織12個と同時期の末梢血8本の提供を受けた。症例1のGVHD皮膚より、患者由来細胞を傷害するがドナー由来細胞を傷害しない2つのアロ反応性T細胞クローンを分離した。これら2つのアロ反応性T細胞クローンのTCRbeta CDR3アミノ酸配列は、その皮膚GVHD組織における頻度第1位および第2位のTCRbeta CDR3アミノ酸配列と同一であり、皮膚浸潤T細胞全体におけるそれら2つの占める割合は98%だった。症例2の皮膚、胃、大腸GVHD組織における頻度第1位のT細胞クローンは、それぞれ組織浸潤T細胞全体の93%、64%、99%を占めており、またそれらT細胞クローンは組織ごとに異なっていた。生検組織中T細胞の多様性をシンプソンの多様性指数(1/Ds)で評価したところ、GVHD組織浸潤T細胞は同時期の末梢血中T細胞と比べて多様性が有意に低かった。グレードIIIのGVHD生検組織はグレードI-IIのGVHD生検組織と比べて組織浸潤T細胞の多様性(1/Ds)が有意に低く、またステロイド抵抗性GVHDの生検組織はステロイド感受性GVHDの生検組織と比べて組織浸潤T細胞の多様性(1/Ds)が有意に低かった。
