Outline of Annual Research Achievements

全身性エリテマトーデス(SLE)の発症にはI型インターフェロン(IFN)が大きく関与しており、インターフェロン制御因子(IRF)は、I型IFNの転写制御に中心的な役割を果たしている。本研究では、SLEの疾患感受性遺伝子であるIRF7とIRF8のSLEの病態における役割について、プリスタン誘発性SLEモデルマウスを用いて検討した。我々は既にIRF7ノックアウト(KO)マウスにプリスタンを投与すると、野生型マウスと同程度に糸球体腎炎を発症したが、自己抗体の産生は認められず、糸球体腎炎の発症と自己抗体の産生の間に解離がみられることを報告した。さらに我々は、自己抗体の産生にはIRF7/I型IFN経路が、糸球体腎炎等の臓器障害にはNF-kB経路が重要な役割を果たしていることを報告した (Miyagawa F et al J Immunol 2016)。一方IRF8KOマウスにプリスタンを投与すると、自己抗体の産生はみられず、糸球体腎炎の程度も野生型マウスよりも軽減するという結果が得られた。IRF7KOマウスおよびIRF8KOマウスの結果より、自己抗体の産生にはI型IFNが重要な役割を果たしていることが明らかとなった。野生型マウス、IRF7KOマウスにおいてはプリスタン投与により腹腔内に炎症性単球が浸潤してくるが、IRF8KOマウスでは浸潤がみられない。さらに炎症性単球はIL-6、TNF-α等の炎症性サイトカインを産生しており、野生型マウス、IRF7KOマウスの腎臓にも浸潤していたことより、炎症性単球がこれらのマウスにおいて腎障害を含めた臓器障害の原因となりうると考えられた。
