再燃前立腺癌への挑戦~脂肪幹細胞ナノDDS技術を駆使した新規テーラーメイド療法~

Outline of Annual Research Achievements |
(目的)体性間葉系幹細胞の一つである脂肪幹細胞(AdSC, Adipose-derived Stem Cell)は、主に再生医療の分野において成果を挙げている一方、近年の研究において制癌作用も併せ持つことが報告されている。本研究では、昨年に続き前立腺癌増殖におけるAdSCの影響を、種々の細胞実験と動物モデルで検討した。 (方法)in vitroにおいて、PC3M-luc2(ルシフェラーゼが導入され高率に転移を来たすPC3)を、AdSCあるいはコントロール(ヒト皮膚線維芽細胞:Normal Human Dermal Fibroblasts (NHDF))と共培養することにより、前立腺癌細胞増殖抑制効果について、Cell proliferation assay、Apoptosis assay、Caspase-3/7 activation assay、microarray analysisを用い検討した。in vivoではヌードマウスに腹腔内移植し、AdSCあるいはNHDFを毎週1回計4回腹腔内投与し、腫瘍の発育をルシフェラーゼアッセイで検討した。 (結果)in vitroあるいはin vivo studyにおいて、AdSC群はNHDF群と比較し、PC3M-luc2の前立腺癌細胞の増殖も抑制し、アポトーシスを亢進させることが示された。
