Outline of Annual Research Achievements

Endometriosis related ovarian neoplasms(ERONs)はエンドメトリオーシスから発生する卵巣悪性腫瘍の総称であり、明細胞癌、類内膜癌、Seromucinous borderline tumorが含まれる。以前、我々はp53変異を基盤とした子宮内膜症の発癌モデルを構築したがマウス移植実験では想定外の結果であった。そこで、新たに子宮内膜症の間質細胞の混入を完全に除外した子宮内膜症不死化細胞EMOsis-CC/TERT10細胞を樹立し、ARID1A変異を基盤としたERONs in vitro発癌モデルを構築する事を目的とした。さらにARID1A変異ERONsに対するARID1Bを標的とした治療戦略についても検討した。子宮内膜症性嚢胞の手術検体を初代培養し、上皮成分だけを分離し、cyclinD1、CDK4、hTERTの遺伝子導入をレンチウイルスベクターにて行った。Pancytokeratin、Calretininのウエスタンブロット法により不死化細胞が上皮細胞由来であるかを確認した。この細胞にCRISPR/Cas9システムを用いてARID1Aの遺伝子ノックアウトを行いEMOsis-CC/TERT10ARID1A-/-細胞のクローニングを行った。リキッドマイクロダイセクション(初代培養+anti-EpiCAM-conjugated beadsとマグネットを用いたセルソーティング)を用いたERONsのmutation解析では19/21(90%)にARID1A変異を認めた。ARID1A遺伝子ノックアウトでは100個のクローンを採取し、3クローンがARID1Aホモ変異であった。EMOsis-CC/TERT10ARID1A-/-細胞でARID1Bをノックダウンすると細胞増殖が抑制された。現在、EMOsis-CC/TERT10ARID1A-/-細胞に変異型K-ras、PIK3CA, c-mycの遺伝子導入を行っており、マウス移植実験で組織型を確認中である。
