Outline of Annual Research Achievements

平成27年度はMyD88の脈絡膜新生血管(CNV)への関与を確認するために、以下の実験を行った。
1.MyD88 inhibitorによるCNVの抑制効果の検証：(1)マウスレーザーCNVを作成後、MyD88 inhibitorを硝子体内に注射した。1週間後に眼球摘出し、フラットマウントを作成、レクチンで染色されたCNVを共焦点顕微鏡で撮影し、ImageJを用いて、CNVの体積を比較した。MyD88 inhibitorはControl inhibitorと比較して有意にCNVの体積を抑制した。
(2)(1)と同様に、注射1週間後にフルオレセイン蛍光眼底造影検査でCNVからの蛍光漏出をグレード分類し、平均値を比較した。MyD88 inhibitorはControl inhibitorと比較して有意にCNVからの蛍光漏出（CNVの活動性）を抑制した。
2.MyD88 inhibitorによる炎症性サイトカインの抑制効果の検証：マウスにレーザーを施行しCNV誘導後、3日後に眼球摘出し、感覚網膜と網膜色素上皮―脈絡膜複合体を分離し、MILLIPLEX Kitsを用いて、マウスサイトカイン・ケモカインを測定した。MCP-1、IL-6、VEGFなどの炎症性サイトカインの過剰発現が認められなかったため、タンパク抽出のtime pointなどを再検討する必要がある。
3.アクリジンオレンジ蛍光染色法を用いたCNV周囲に集積する白血球数の比較：レーザーCNV作成後、1週間後にアクリジンオレンジ蛍光染色法を用いてCNV周囲に集積する白血球数がMyD88 inhibitorにより抑制されるか対象と比較した。MyD88 inhibitor投与群では対象と比較して有意に集積した白血球数が少なかった。

Strategy for Future Research Activity

次年度からはMyD88を介した細胞内シグナル伝達機構の解明と、CNVにおける炎症とアポトーシスの関与解明を目指す。

Causes of Carryover

消耗品などの価格で端数が生じたため、27,967円の差額が生じた。
