性的二型核に着目した脳への系統進化的アプローチ

Outline of Annual Research Achievements

性差がみられる神経核（性的二型核）は様々な動物の脳に存在する。しかし、性的二型核の種差に関する研究知見は乏しい。マウスの脳には、雄優位な性的二型核である分界条床核とカルビンディンSDN、そして、分界条床核とカルビンディンSDNの間にみられる雌優位なサンドイッチ領域がある。本研究では、これらの性的二型核が他の動物にも存在する否か明らかすることを目的にしている。当該年度は、ウズラとスンクスを対象にして、組織学的解析を行った。マウスの分界条床核とカルビンディンSDNのマーカーであるカルビンディンの免疫組織化学の解析から、スンクスにはマウスのカルビンディンSDNに相同する性的二型核が存在することが明らかになった。また、カルビンディンを発現しない細胞から構成される性的二型核がスンクスの分界条床核にあることも分かった。しかし、ウズラの脳では、マウスのカルビンディンSDNおよび分界条床核に相当する領域を見出すことはできなかった。サンドイッチ領域の同定に有効なマーカーを探索することを目的として、雌雄マウスのサンドイッチ領域より得たRNAサンプルを用いた網羅的遺伝子発現解析を行った。その結果、発現量に性差がみとめられる幾つかの遺伝子（神経ペプチド、ノンコーディングRNAなど）が見つかった。
