Outline of Annual Research Achievements

二光子顕微鏡を用いて麻酔下のGFAP-eGFPマウス大脳皮質におけるアストロサイトのin vivo imagingを行い,同時にアストロサイト足突起へのレーザーアブレーション(ごく微小な領域にレーザーを照射して損傷)も行った.足突起をアブレーションした後に継続的に損傷部位の観察を行った結果,失われた足突起はおおよそ24時間以内に血管に沿って伸長して再接触が認められた.この再接触は,アブレーションしたアストロサイトの生死や損傷部位に関係なく引き起こされていた.このことは,アストロサイト足突起が脳血管を覆っている状態は,生体にとって生理学的な意義があることを示唆している.
さらに,アブレーションによってアストロサイトの足突起が脳血管から解離しても,血管を標識したエバンスブルー(EB)の漏出は見られなかった.また,より分子量が小さいFITCで血管を標識した際にも血管からの漏出が認められなかった.このことは,アストロサイトの足突起は血液脳関門(BBB)に対して直接的な機能は持っていない可能性を見出した.
アストロサイトの足突起を損傷してもBBBの破綻は認められなかったが,その一方で,脳血管をアブレーションした際にはBBBの破綻によるEBの漏出が損傷後1分以内に観察された.このことから,アストロサイトの足突起はBBBにおいて物理的な障壁として直接は機能してはいない可能性が明らかとなった.
