Outline of Final Research Achievements |
We planed to establish the new experimental system to analyze disease-associated single nucleotide polymorphisms (SNPs) using a reverse genetics technique. In this study, we developed a method to generate SNP model cells using CRISPR/Cas9 system. This method has enabled us to carry out comparative study of candidate SNP in human cultured cells under genetically uniform backgrounds. The SNP model cell lines we established in this study, which have either mono- or bi-allelic mutation, showed increased chromosome instability.
