Outline of Annual Research Achievements

In this study, we attempted to identify the microbes symbiont in the wood wasp from Hokkaido Forest. We were able to capture two wood wasp species, Sirex nitobei, and Xiphydria camelus. One bacteria and one fungus were isolated from a mycangia of X. camelus, and were determined to be Rahnella spp., and Daldinia decipiens species by 16S and ITS sequencing, respectively. Furthermore, we determined the hemicellulose-degrading activity in the supernatant when D. decipiens was cultured in the liquid medium containing polysaccharides such as cellulose, hemicellulose and wood biomass as sole carbon source.

Reason

Capturing the woodwasps from native wood is not trivial task. We are hoping to collect more woodwasp insects in this year (2016) in order to increase sample numbers and number of microbes from wood devastating insects to determine their cellulolytic capability.

Strategy for Future Research Activity

The Biomass-degrading potential in the secretome from D. dicipiens was determined. We will be identifying the enzymes that degrade biomass by secretome analysis.
In order to statistically analyze the wood wasp symbionts, we will capture more wood wasp insects from the Hokkaido forest.

Causes of Carryover

We captured wood devastating insects less than what we expected, and the next generation sequencing analyses were not carried out. Thus, we needed to carry over the funding to the next fiscal year for upcoming insect-microbe analyses.
