Characterization of the glycosylation pattern in cholangiolocellular carcinoma

Outline of Annual Research Achievements

【最終年度に実施した研究の成果の概要】細胆管細胞癌 20症例、末梢型肝内胆管癌 26症例、肝門周囲型肝内胆管癌 25症例、肝細胞癌 25症例の手術標本に対して免疫組織染色を実施し、糖鎖およびコアタンパク質の発現パターンの解析を行なったところ、細胆管細胞癌において硫酸化糖鎖とepithelial membrane antigen（EMA）が細胆管用様構造の内腔面に共発現していることを明らかにした。細胆管細胞癌における硫酸化糖鎖の生合成に関与する硫酸化糖鎖合成酵素を明らかにするため、肝内胆管癌由来の細胞株にGlcNAc6ST-1またはGlcNAc6ST-2のcDNAを遺伝子導入したところ、GlcNAc6ST-2の存在下で硫酸化糖鎖が細胞膜に発現した。野生型およびGlcNAc6ST-1ノックアウトマウスでは、小径肝内胆管で硫酸化糖鎖の発現がみられたが、GlcNAc6ST-1およびGlcNAc6ST-2のダブルノックアウトマウスではその発現が消失していた。【研究機関全体を通じて実施した研究の成果の概要】本研究では肝原発性悪性腫瘍の中でも比較的まれで、発症機構が明らかにされていない細胆管細胞癌に焦点を当て、他の肝原発性悪性腫瘍とは異なる発現パターンを示すEMAの糖鎖構造を明らかにすることを目的とし、1）細胆管細胞癌の細胆管様構造では内腔面に硫酸化糖鎖とEMAが共発現していること、2）EMAが硫酸化糖鎖のコアタンパク質となること、3）細胆管細胞癌における硫酸化糖鎖の生合成にGlcNAc6ST-2が関与していることを明らかにした。これらの研究成果は、細胆管様構造の内腔面における硫酸化糖鎖の発現が細胆管細胞癌の病理組織学的診断におけるマーカーとして利用可能であることを示している。
