Research Abstract

Cell surface markers such as CD34 expressed on the surface of the cell are useful for identifying various stem cells, and then, material surfaces where the antibodies against CD34 molecules are immobilized would possess specific potential against hematopoietic stem cells or EPCs. In the present research, novel biomaterials for tissue regeneration were developed based on the specific interaction between the immobilized bio-active molecules and stem cells. First of all, immobilizing method of antibodies onto poly(lactic acid) materials has been developed and the porous tubular scaffolds for regenerative blood vessels with stem cell specific surfaced were designed. The CD34 positive stem cells derived from the bone marrow cells were selectively adsorbed on the these active surface and, suggesting these surface-activated scaffolds were useful as the artificial blood vessels. In addition, the subpopulation of the stem cell was newly found being able to be separated on the above-mentioned antibody-immobilized surfaces. This phenomenon is very useful in the field of tissue regeneration since it is quite difficult to purify only the expected stem cells. We established novel cell separation column system based on this cell rolling phenomenon on the antibody immobilized surfaces, and successfully isolated the fraction that efficiently differentiate to the target cells.
The other way for surface modification of the poly(lactic acid) scaffolds was to use newly synthesized amphiphilic block copolymers composed of the oligopeptides and oligo(lactic acid) s. When the nerve guide tubes made of poly(lactic acid) were modified with theseoligo(lactic acid) -IKVAV peptide amphiphilic block copolymer, the enhanced nerve regeneration had been observed in vitro and in vivo.
