Research Abstract |
To investigate the influence of gene polymorphism of a homocysteine metabolism-related enzyme, methylenetetrahydrofolate reductase (MTHFR), on physiological polymorphism of arteriosclerosis, MTHFR genotypes were analyzed, and the degree of progression of arteriosclerosis was determined by measuring the intima/media complex thickness (IMT) and pulse wave velocity (PWV). The importance of inflammatory reaction involved in the progression process of arteriosclerosis has recently been reported, and serum high-sensitive CRP (hs-CRP) has been identified as an independent risk factor of angiopathy by epidemiological studies. We measured hs-CRP as a systemic factor involved in the progression of arteriosclerosis, in addition to serum biochemical testing including serum lipid measurement. Sex differences in the levels of hs-CRP, the currently most sensitive index of changes in the arteriosclerosis course, and plasma folate were already present in adolescence, and the carotid intima/media thickness (CIMT) measured by cervical ultrasonography thickened with aging, increasing the sex difference (Hara et al., Clin Chem Lab Med 2006). We have recently analyzed homocysteine-and CIMT-determining factors in middle-aged to elderly Japanese, and found that systolic blood pressure is an independent factor, in addition to aging and gender (Takamura et al., submitted). These findings indicate the importance of control of blood pressure, particularly, systolic blood pressure, for the prevention of arteriosclerosis. Analysis of the influence of MTHFR gene polymorphism on physiological polymorphism of arteriosclerosis based on the previous study data is underway.
