Research Abstract

1.Production of PAU (Polyaminourethane)-coated hydrophilic small caliber regenerative graft and creation of canine model for judging the effectiveness of this new graft : 3mm diameter graft was coated and used for canine cervical artery implantation model, but unfortunately few patent grafts were obtained for evaluation due to intimal hyperplasia of anastomotic site. The patent grafts ware used for evaluation. The PAU-coated graft was histotropic macroscopically and inner-layer of it was covered with less thrombus and endothelial-like cell microscopically.
2.Determination of optimal conditions of bone marrow derived endothelial progenitor cell adhesion and proliferation on "PTFE (Polytetrafluoroethylene) sheets" coated with PAU, ECM (extracellular matrix), and growth factors in vitro : G-CSF mobilized peripheral-blood mononuclear cells were used for determination of PAU/ECM combinations on "PTFE sheets" in culture plates. Six ECM, type I & IV Collagen, Fibronectin, 2 Fibronectin peptide and Vitronectin were used for making PAU/ECM combinations. Among PAU/ECM combinations, "type I Collagen+Fibronectin-GRGD peptide+Vitronectin" was most efficient, and "Fibronectin+Vitronectin" was next for G-CSF mobilized peripheral-blood mononuclear cell proliferation on "PTFE sheets".
3.Evaluation of the new small caliber regenerative graft coated with PAU/ECM combination based on the data of experiment "2" : It remains to be unclear.
