Research Abstract |
The researchers have identified that Valosin-containing protein (VCP) to be correlated with the metastasis of a murine osteosarcoma cell line by using the mRNA subtraction technique. This project was designed to identify whether or not VCP expression level in cancer cells was correlated with the rate of recurrence and prognosis of patients with cancer and clarify the transcriptional mechanism of VCP. The result showed that VCP expression is correlated with the progression and prognosis of cancers arising from lung, prostate, pancreatic endocrine, esophagus, gingiva, and thyroid. Oncol. 2005;12:925-934) Transcriptional activity of VCP gene was analyzed by Luciferase reporter assay. Luciferase reporter assay revealed two candidate transcription factors, PBX1 and ELF2 which might be important in VCP transcription. Chromatin immunoprecipitation assay revealed the binding of PBX1 and ELF2 to the 5' flanking region DNA sequence. Cellular knock-down of PBX1 and ELF2 expression with siRNA transfection led decrease and increase of VCP expression, respectively. These findings indicate that PBX1 and ELF2 are important transcription factors for VCP expression.
