Research Abstract

In rats which serum corticosterone concentration was adjusted in the infant stage, time-courses of hippocampus NMDA receptor expression at the stress-loading condition and performance against stresses were compared with control subjects. Further, protective effects of antagonists of NMDA receptor were studied. SD rats were assorted into four groups ; the group of control subjects, the group with adrenorectomy, the group with subcutaneous administration of corticosterone after adrenorectomy, and the group with subcutaneous administration of corticosterone without adrenorectomy. Concentrations of serum corticosterone in all of those rats were adjusted. Each group was divided into two subgroup ; with administration of memantine using osmotic pomp and without that. The serum concentration of corticosterone in ach rat was monitored. Two weeks after those procedures, the learning deficit in Y maze was measured at day 1, day 5 and day 7, and the amount of activity was also measured by Animex using infrared rays. Consequently, any differences in body weight and total activity was seen among those groups. However, apparent learning deficit was found in the group with subcutaneous administration of corticosterone after adrenorectomy. And, in the subgroup with memantine in the group with subcutaneous administration of corticosterone without adrenorectomy, learning deficit was not seen, and decrease of total activity was seen.
Using in situ hybridization, mRNA expressions of NMDA receptor subtypes were analyzed in the group with subcutaneous administration of corticosterone after adrenorectomy. Expression of NR2B tended to be decreased, suggesting that corticosterone might influence on learning ability via N2RB.
