| Research Abstract |
癌のリンパ節転移のメカニズムは複雑であるがおよそ以下の順によるメカニズムでリンパ節転移がしょうずるのではないかと考えられる。(1)腫瘍の増殖(2)リンパ管新生能のあるサイトカインの分泌(3)リンパ管新生(4)腫瘍細胞の毛細リンパ管への移動と浸潤(5)腫瘍細胞のリンパ管腔内への遊走と滑り込み(6)リンパ管内でのリンパ流の乗った腫瘍細胞のsentinel nodeへの移動(7)sentinel node皮質への浸潤(8)さらに他のリンパ節への転移。肺癌リンパ節の転移のバイオマーカーによる検索の本研究において(2)のリンパ管新生能のバイオマーカーにおいて従来からのVEGF-C、VEGF-Dの他にAngiopoietin-2もまた肺癌リンパ節転移のバイオマーカーであることを本研究者は英文誌に発表した。Overwxpression of Ang-2 mRNA in non-amall cell lung cancer : Association with angiogenesis and poor prognosis. Oncol Reports 12:849-853, 2004.である。(4)腫瘍細胞の毛細リンパ管への移動と浸潤においては、腫瘍が細胞間マトリックスに浸潤するにおいて本研究者はIntegrin-linked kinaseに注目し英文誌に発表した。Increased expression of integrin-linked kinase is associated with shorter survival in-non-small cell lung cancer. BMC Cancer 5:1, 2005.である。また、この時腫瘍内圧が大きく関与することが知られているが、その関与の物質としてG-protein inwardly rectifying potassium channel 1に注目しこれを本研究者は英文誌に発表した。G-Protein inwardly rectifying potassium channel 1(GIRK1) gene expression correlates with tumor progression in non-small cell lung cancer. BMC Cancer 4:79, 2004.である。(5)腫瘍細胞のリンパ管腔内への遊走と滑り込みに関して本研究者はAutocrine motility factor receptorに注目し英文誌に発表した。Significance of autocrine motility factor receptor gene expression as a prognostic factor in non-small-cell lung cancer. Int J Cancer 95:384-389, 2001.である。(6)リンパ管内でのリンパ流の乗った腫瘍細胞のsentinel nodeへの移動に関して本研究者はOverexpression of CCR7 mRNA in non-small cell lung cancer : Correlation with lymph node metastasis. Int J Cancer 105:186-189, 2003.に発表した。
