Research Abstract

Objective
The aim of the present study is to elucidate the efficacy of granulocyte-colony stimulating factor (G-SCF) to treat spinal cord injury.
Materials and Methods
Bone marrow cells were isolated from male GFP Tg mice and transplanted intravenously into tail vein of lethally irradiated female C57BL/6 mice. Four weeks after bone marrow transplantation, spinal cord injury was produced by a static load (20g,5min.) at T8 level. G-CSF (200μg/kg/d) was injected subcutaneously for 5 days. Hind limb motor recovery was assessed with Motor Function Scale (Farooque,2001). Spinal cord section was made for histology. Immunohistochemistry for GFP and cell lineage markers was performed to evaluate mobilization of bone marrow-derived cells into injured spinal cord.
Results and Discussion
G-CSF treated mice showed significant recovery of hind limb function compared to that of the control mice. Immunohistochemistry revealed that GFP-positive cells were detected in the spinal cord sections, indicating that bone marrow-derived cells were mobilized and survived within the injured spinal cord. In conclusion, G-CSF may have beneficial effect for spinal cord repair.
