Research Abstract |
Rheumatoid arthritis is associated with the citrullination. Autoantibodies to the citrullinated peptides derived from filaggrin or fibrin are specifically produced in the disease. The pathological involvement of autoantibodies is, however, still unclear since a filaggrin or a fibrin is not a major or a specific molecule in joints. On the other hand, a type II collagen is a major and a specific molecule in articular cartilage. Thus, the citrullination of type II collagen and the production of autoantibodies to the citrullinated type II collagen were analyzed in the present study. The autoantibodies to the citrullinated type II collagen were positive in 78.5% of sera from 130 patients with rheumatoid arthritis. One serum was positive for autoantibodies in 31 patients with systemic lupus erythematodes and two sera were positive in 55 patients with osteoarthritis of knee joints. litis, 10 psoriatic arthritis and 8 systemic sclerosis. Anti-citrullinated type II collagen antibodies and fragments of citrullinated type II collagen were found in the synovial fluids obtained from affected knee joints of 15 patients with rheumatoid arthritis. Moreover, anti-citrullinated type II collagen antibodies were isolated from the synovium of affected knee joints in 8 rheumatoid arthritis by the method using the dissociation buffer of antigen/antibody immunocomplex, but not by using a standard buffer. These findings indicate that autoantibodies react with citrullinated type II collagen are specifically produced and immunocomplex conformed with fragments of citrullinated type II collagens and binding autoantibodies was deposited at the inflamed articular synovium in rheumatoid arthritis. The assay for anti-citrullinated type II collagen antibodies may be significant for diagnosis of rheumatoid arthritis and the deposition of immunocomplex at the articular synovium may be involved in the pathogenesis of the disease.
