Research Abstract |
THERE IS GROWING EVIDENCE THAT EPIGENETIC CHANGE OF GENOMIC DNA PLAYS AN IMPORTANT ROLE FOR TISSUE-SPECIFIC MOLECULAR EXPRESSION. SINCE THESE CHANGES ARE EASILY CONTROLLED BY SEVERAL ARTIFICIAL FACTORS INCLUDING ENVIRONMENTAL FACTOR< THE RELATIONSHIP BETWEEN THE EPIGENETIC CHANGE OF GENOME AND THE LIFESTYLE-RELATED ILLNESSES ARE FOCUSED ON CLINICALLY. THE PRIMARY OBJECTIVE OF THIS RESEARCH IS TO EXAMINE THE EPIGENETIC EFFECT OF ARTIFICIAL REPRODUCTIVE TECHNIQUE(S) ON GAMETE/ZYGOTE AND EARLY EMBRYONAL DEVELOPMENT. AS AN INITIAL STEP< WE TRIED TO ESTABLISH A SINGLE CELL PCR TECHNIQUE, BECAUSE AT MOST 20-30 EGGS ARE RECOVERED FROM AN ADULT MOUSE WITHOUT TREATMENT OF SUPEROVULATION. SUBSEQUENTLY, WE CHARACTERIZED TEX101 GENE, RECENTLY IDENTIFIED AS A GERM-CELL-SPECIFIC MARKER, FOR A CANDIDATE OF THE TARGET GENE SHOWING EPIGENETIC CHANGE AFTER ENVIRONMENTAL EXPOSURE. DELETION ANALYSIS OF THE CHIMERIC STRUCTURES INDICATED THAT SEQUENCES ESSENTIAL TO GENE EXPRESSION ARE PRESENT ON THE 5'-FLANKING REGION BETWEEN 3186 AND +14, WHERE THE CLUSTER OF FIVE CAAT BOXES IS LOCATED. THESE FINDINGS SHOULD FACILITATE AN UNDERSTANDING OF THE REGULATION OF TEX 101 EXPRESSION DURING GAMETOGENESIS, AND MAY BE USEFUL FOR FURTHER STUDIES OF THE GENE INCLUDING EPIGENETIC ANALYSIS.
