DNA合成酵素新規分子種選択的阻害物質の探索・分子設計とケミカルノックアウト解析

Research Abstract

昨年度の新規DNA合成酵素(DNAポリメラーゼ、pol)分子種群に対する特異的阻害物質のスクリーニングにより、食品素材である「ふきのとう」から単離・精製したフェノール系化合物であるPetasiphenolがpolλを特異的に阻害することを見出した。Petasiphenolと同じフェノール系化合物であり、化学構造が類似しているクルクミン(Curcumin)にもpolλ特異的阻害活性があった。
そこで、クルクミンをリード化合物として、クルクミンの各種誘導体13物質を有機化学合成した。これら13物質のpol阻害活性を評価した結果、Monoacetyl curcuminがクルクミンよりも強いpolλ阻害活性を示した(Monoacetyl curcuminのIC_<50>値は3.9μM、クルクミンのIC_<50>値は7.8μM)。
Monoacetyl curcuminのpolλ阻害活性について、表面プラズモン共鳴センサー装置(BIAcore X)および分子量解析装置(MALDI-TOF MS)を用いて解析したところ、Monoacetyl curcuminは基質である二本鎖DNAに結合するのではなく、polλに結合することによって阻害することがわかった。さらにpolλはN-末側に18kDaのBRCTドメインを有しており、Monoacetyl curcuminはBRCTドメインに結合した。
以上の結果を踏まえて、Monoacetyl curcuminとpolλ・BRCTドメインのコンピュータ・シミュレーションにより結合(ドッキング)モデリングを実施した。Monoacetyl curcuminのアセチル基とBRCTドメインのLys63が水素結合することが示唆された。
