Outline of Annual Research Achievements

本研究では、近年、国際社会において大きな関心を集めている新規POPおよび代替物質を対象に、アジア太平洋地域の環境・生態系汚染の現状と経年変化およびバイオアッセイ等による影響評価の基礎データを集積・解析し、環境改善や対策技術構築のための科学的根拠を提示することを目的とする。初年度は、臭素系の代替難燃剤であるデカブロモジフェニルエタン(DBDPE)やビストリブロモフェノキシエタン(BTBPE)、そして塩素化難燃剤であるデクロランプラス(DPs)の分析法を確立し野生魚類と鯨類の組織試料に適用した結果、DBDPEおよびBTBPEは検出下限値以下であったが、魚類の筋肉組織からDPsが検出された。興味深いことに、インド、インドネシア、フィリピンで採取した魚類のDPs濃度は検出下限値レベルであったが、日本とベトナムの検体は相対的に高い濃度を示した。また、日本で採取した魚類ではHBCDsの高濃度蓄積も認められ、これらハロゲン化難燃剤による水域汚染の顕在化が示唆された。代替ハロゲン化難燃剤の検出は認められなかったが、日本沿岸に座礁したネズミイルカとイシイルカから新規POPsが検出され、とくに外洋性種であるイシイルカのHBCDs濃度はレガシーPOPsと比べても相対的に高値を示した。愛媛大学沿岸環境科学研究センターのes-BANKに冷凍保存された1980年代以降の検体を化学分析し経年変化を解析した結果、イシイルカのHBCDs濃度は明らかに増加していることが判明した。また、イシイルカではPBDEs濃度も有意に増加していた一方で、ネズミイルカのBFRs濃度は明らかな変動を示さなかった。これらの結果を考慮すると、新規POPsであるPBDEsとHBCDsによる沖合への移動拡散は継続していると推察される。

Reason

初年度は、PBDEsの代替難燃剤であるデカブロモジフェニルエタン (DBDPE) やビストリブロモフェノキシエタン (BTBPE) に加え、塩素化難燃剤であるデクロランプラス (DPs)について、臓器試料に適用可能な分析法を確立することができた。リン酸エステル系難燃剤 (PFRs) についても分析機器の条件設定は最適化できたが、臓器に由来するマトリックスの除去が今後の課題となった。代替ハロゲン化難燃剤の分析法を用いて日本、インド、インドネシア、ベトナム、フィリピンで採取された魚類の汚染実態を調査した結果、DBDPEとBTBPEは検出下限値以下であったが、日本とベトナムの検体からDPsが検出され、途上国においても汚染が顕在化していることが判明した。また新規POPsであるHBCDsについては、日本の魚類で他の国より明らかに高い濃度の蓄積が認められ、沿岸性および外洋性の鯨類組織中でもレガシーPOPsに匹敵する濃度の検出が明らかとなったことから、相対的に高いレベルの曝露が日本に生息する野生生物へ及んでいることが推察された。とくに、外洋性種である鯨類のHBCDs濃度は1980年代以降急激に上昇し、2000年以降も有意な増加傾向を示していることを初めて見出し、外洋の汚染が長期化する可能性を提示することができた。現在、新規POPsと代替物質の発生・集積源が遍在すると考えられる途上国の分析を進めており、多様な動物種を対象に生物蓄積の解明と汚染の過去復元・将来予測に関する研究も継続しデータを集積する計画である。

Strategy for Future Research Activity

代替難燃剤であるリン酸エステル系難燃剤(PFRs)の前処理法を改良し、生体試料に適用可能な分析法の確立を目指す。また、多様な動物種を対象に、新規POPsおよびPOPs代替物質の汚染実態と蓄積特性の解明を試みる。さらに、これら汚染物質の主要な発生・集積源と考えられる途上国の汚染と網羅分析によるスクリーニングを展開する。平成29年度における課題の詳細は以下の通り。
１．分析法の開発：　初年度に、生体試料に適用可能なDBDPEやBTBPEなどの代替臭素化難燃剤に加え、塩素化難燃剤であるDPsの分析法を確立したが、PFRsについては生体組織に由来するマトリックスの除去が不可欠となっている。そこで、異なる担体を有する3種類のカートリッジを用いた前処理法を検討し、高精度分析法を確立する。
２．生物蓄積性の解明：　鯨類に加え、猛禽類などの野生鳥類における新規POPsとPOPs代替物質の蓄積特性を解明する。また、PBDEs の高濃度曝露による健康影響が危惧されている室内ペットにも着目し、POPs 関連物質の曝露実態を明らかにする。とくに甲状腺ホルモン恒常性の異常が多く観察されるネコについては、その原因物質として疑われている代謝物についても分析をおこなう。
３．廃棄物処理場の汚染実態と網羅的スクリーニング：　PBDEsの主要な環境放出源となっている途上国の電子・電気機器廃棄物(e-waste)処理場の調査を実施する。作業環境の室内ダストを分析し、新規POPsおよび代替物質の汚染実態を明らかにする。また、魚介類などについても調査し、ヒトに対する曝露源や曝露ルートを解明する。さらに、2次元ガスクロマトグラフ飛行時間型高分解能質量分析計を用いた網羅分析に着手する。
