Outline of Annual Research Achievements

本研究は,プラズマが生成する電荷刺激による細胞応答を明らかにすることで,電荷と細胞の相互作用機構を明らかにし,新しい学理である電荷細胞学の基礎となる学理の解明を目的とする.
平成30年度は,平成29年度に作製した電荷刺激を細胞に与える装置を利用して,詳細な電荷刺激に対する細胞応答の解析を進めた.電荷刺激は,プラズマ発生用のパルス電源を改良し,電流の立ち下がりを早めるためにバイパス抵抗と保護抵抗の比を変更した装置で与えた.電荷刺激は電流パルスとして与え,最大電流200-220 mAで印加時間は200 ns程度,100 Hz,印加電圧990V-1000Vの条件で与えた.塩橋を介すことで化学種の影響を排除し,電荷刺激印加中のpH値と温度は,電流パルスを印加しない場合と概ね同じ値となることを確認した.細胞応答については,形状,生存率,遊走性,増殖性について検討した.電荷刺激による生存率の変化は見られなかったが,細胞形状が大きくなる現象が観察された.運動能を確認するために,細胞骨格を担うアクチンフィラメントとチューブリンの状態を蛍光染色により観察したところ,アクチンフィラメントのみが進展し,細胞形状が大きくなることを明らかにした.この成果は,Journal Physics D: Applied Physicsに掲載された.また,平成29年度に取り組んだ指向性マイクロジェットの発生原理に関わる研究についても,平成30年度に成果を取りまとめ,Applied Physics Lettersに掲載した.さらに,平成28年度から取り組んできた,細胞への電荷直接刺激法の一つとして極微小ストリーマ生成で優位性のある負極性ストリーマの進展・分岐機構の成果も取りまとめ,Journal of Applied Physicsに掲載した.
