Outline of Annual Research Achievements

1.ホヤVSPの分子特性から推定されてきた、イノシトールリン脂質を基質とする4つの酵素反応のうち、どの過程が精子でのVSP分子機能に重要であるかを明らかにするため、東京医科歯科大学佐々木雄彦博士、秋田大学中西博士らが開発したイノシトール環に結合する部位の異なるイノシトールリン脂質を区別できる質量分析手法によりVSPノックアウトマウスとヘテロマウスの間で精子でのイノシトールリン脂質種のプロファイルを比較した。その結果VSPノックアウトマウス由来精子では、有意にPI(4)Pのレベルが低下しており、その一方PI(4,5)P2のレベルが増加していた。このことからマウス精子のVSPはPI(4,5)P2の5位のリン酸を脱リン酸化する活性をその生理機能とすることが明らかになった。
2.前年度までに明らかになったVSPノックアウトマウスにおけるSLO3活性を電気生理学的計測により解析した。精子からのホールセルパッチクランプ法によりSLO3電流をClofilium感受性電流として定量したところノックアウトマウス由来精子において有意にSLO3電流が増加しており、尾部でのCa2+流入がVSPノックアウトマウスでは増加していることと尾部にSLO3が存在することと考え合わせると、SLO3活性の亢進によりCa2+流入のドライビングフォースが増加したために細胞内Ca2+濃度が増加して運動性の異常に繋がったと考えられた。
3.このSLO3電流の亢進がPI(4,5)P2レベルとどのように関係するかを調べるため、パッチ電極内に抗PI(4,5)P2抗体を含んだ状態でノックアウトマウスとヘテロマウスの電流を比較したところ、両者に電流量に差は見られなかった。これによりSLO3電流の活性化はVSP活性が失われたことによるPI(4,5)P2のレベルの増加によりもたらされると結論した。
