| Outline of Annual Research Achievements |
Wistarラットに20㎎/㎏のアクリルアミドを1日1回、週7日、5週間連日投与した。脳各部位のホモジェネートから酸による除タンパクを行い、モノアミンレベルを高速液体クロマトグラフィーによって定量した。その結果、ノルアドレナリンレベルが減少しているものの、セロトニンレベルには変化がないことが明らかとなった。また一部の脳部位ではドーパミンレベルの上昇が見られた。さらにドーパミンベータハイドロキシラーゼおよびセロトニントランスポーターに対する抗体を用いた免疫組織化学では、ドーパミンベータハイドロキシラーゼ陽性の神経軸索密度のみが減少し、アクリルアミド曝露によるノルアドレナリン神経特異的な影響が観察された。次にC57BL/6Jマウスにアクリルアミド20㎎/㎏を腹腔投与し、1時間後、3時間後、6時間後に左心室から4％パラフォルムアルデヒド緩衝液を用いて灌流固定を行った。灌流終了1時間経過後、脳を剖出し、24時間、4％パラフォルムアルデヒド緩衝液にて後固定を行った後、10、20、30パーセントスクロースに順次置換し、凍結ブロックを作成した。凍結標本を用いてLC3B抗体で免疫染色を行った。その結果、小脳プルキンエ細胞におけるLC3Bタンパクの発現はアクリルアミド投与後1時間が最も高く、次いで3時間後に見られ、6時間後には消失していた。本研究は、ソフトな親電子性物質アクリルアミドが小脳プルキンエ細胞においてオートファゴゾームを一過性に形成することが明らかとなった。

LC3遺伝子改変動物を用いた実験を準備しているが、児の生存が悪く、繁殖に支障が生じている。現在、飼育上の問題を検討中である。

| Strategy for Future Research Activity |
小脳プルキンエ細胞におけるオートファジーを確認するためには電子顕微鏡による観察が必要である。免疫組織化学と電子顕微鏡観察が両立する固定液選択のための実験を行ったうえで、古典的な電子顕微鏡観察および免疫電子顕微鏡観察を行う。培養細胞によるオートファジーメカニズム解明もあわせて進める。
