Outline of Annual Research Achievements

生命現象の解明を目指した高精度な細胞機能制御技術の開発は、生命科学研究における究極の目的の一つである。なかでも光を活用した細胞機能制御技術は、簡便性や細胞応答性の高さから注目を集めている。しかし、従来技術は、生体透過性の低い光を利用するため生体深部領域の細胞機能を制御することはできない。また、安全性の低いウイルスを利用するため医療応用が難しい。従って、本研究目的では、生体透過性の高い近赤外光によって発熱可能なカーボンナノチューブと特定の温度で内包分子を放出する温度感受性リポソームを組み合せることで、ウイルスフリーで、かつ生体深部の細胞機能をナノメーターレベルで光により制御する技術を構築する。本研究は、とりわけガンや神経変性疾患に対する新しい分子標的医薬や先進医療技術のための普遍的な技術となる。本研究では、最終構想にマウス生体中の細胞内酵素反応制御が可能な機能性ナノ複合体の開発を掲げており、本最終構想を達成するために、本年度は、まず、コア技術となる細胞・組織集積能を強化したナノ複合体の開発を目指した。

Strategy for Future Research Activity

作製した機能性ナノ複合体を細胞内に導入し、800 nmの近赤外レーザーを照射することで酵素反応制御効果を検証し、最大の蛍光強度変化量が得られる最適条件を探索する。
