Outline of Final Research Achievements

In this study, we developed microdevices for use in vascular bioassays. We have developed a device that mimics blood flow and can be used for fluid culture, a cell-stretching device that mimics vasodilation, and a device made of gelatin. By changing the cells to be cultured in these devices, various vascular models can be created. Applications include: 1) hematopoiesis during early mammalian development, and 2) smooth muscle cell proliferation in pulmonary hypertension.

Academic Significance and Societal Importance of the Research Achievements

細胞バイオアッセイは、動物を使わずに物質が生物に及ぼす影響を細胞で評価する分析方法である。近年、ヒトの細胞微小環境を正確に反映する組織モデルによるアッセイの開発が求められている。本研究では、血管内皮細胞の培養条件を詳細に調べ周囲の細胞や間質とともに再構成することで、生理的に適切な血管モデルの構築を試みた。開発したモデルは発生学から難治疾患解明まで広く血管研究に貢献できる。
