横波超音波の固有振動数変動による抗原抗体反応の高感度質量計測

Outline of Annual Research Achievements

本研究は、液体中で分子間の相互作用を検出できる高感度センサの実現を目指すものである。具体的には、独自の傾斜配向ScAlN膜を用いた擬似横波薄膜共振子を用いて、液体中で持続的共振を実現し、共振周波数変動を用いて、分子間相互作用の検出を目指している。
計画通り、傾斜配向ScAlN膜の傾斜角度、配向度を極点X線回折法により評価して、成長条件を最適化することができた。これにより、傾斜角度30度以上、χ走査ロッキングカーブ半値幅15°以下の傾斜配向薄膜が再現性良く成長できるようになっている。さらに成長させた薄膜を用いて、横波薄膜共振子を作製し、恒温槽を用いた液中測定システムを構築した。粘性液体(グリセリン水溶液)をセンサに付加し、その粘性侵入長を質量負荷として模擬し、粘性を変化させた際の共振周波数変化を上記システムにより測定した。その結果、粘性変化に応じた妥当な共振周波数の変化を観測することができた。
また計画通り、極性反転構造によるセンサ面積拡大を目指して、傾斜反転構造が成長できるよう、真空チャンバーに回転導入器を設置する改造を行い、4層以上のc軸傾斜反転ジグザグ構造を作製にも成功した。この構造の高次モード共振特性をネットワークアナライザを用いた高周波測定系により評価するとともに、電気機械結合係数を測定した。c軸傾斜反転ジグザグ構造に基づく極性反転構造を考慮したMasonの等価回路モデルを構築し、高次モード横波共振特性をシミュレーションした結果と、上記の実測の高次モード共振特性を比較した結果、よく一致していることを確認するとともに、センサ面積の拡大も確認できた。

Reason

恒温槽を用いた液中測定システムを構築し、粘性を変化させた際の共振周波数変化を測定した結果、粘性変化に応じた妥当な共振周波数の変化を観測することができた。

Strategy for Future Research Activity

今後、計画どおり500MHz~800Mzの音響多層膜を用いたた傾斜配向ScAlN膜の擬似横波共振子を作製する。これを用いて、生体高分子を用いた分子間相互作用の検出実験を行い、高感度センサの実現を目指していく。
