家畜対応型の抗病性イムノバオティクス選抜・評価系の確立とその応用戦略

Outline of Annual Research Achievements

ブタやウシなどの家畜は、幼若期に腸管感染症が多発し、成長遅延から生産性の著しい低下を招き産業被害が増大している。その対策として多用される抗菌剤により薬剤耐性菌の出現リスクが高まり、家畜やヒトへの新たな健康危害リスクの増加が懸念されている。そのため、抗菌剤のみに頼らない家畜の健全育成技術の開発が急務の課題とされている。本研究は、世界初となる家畜対応型イムノバイオティクスのインビトロ抗病性選抜・評価系を構築し、その発展的利用による家畜健全育成技術基盤の確立を目指した。本年度は、昨年度までに構築したロタウイルスを用いたウイルス感染・評価系により、抗病性イムノバイオティクスの選抜・評価をさらに進め、子ブタへの投与によるインビボ検証を開始すると共に、イムノバイオティクス評価系の発展的構築による抗病性機構解明を目指した。具体的には、以下の研究成果が得られた。
１．抗病性イムノバイオティクスの選抜・評価：前年度からの継続で、乳酸菌やビフィズス菌を中心としてロタウイルスや２本鎖RNAを用いた評価により、抗病性イムノバイオティクス候補菌株を選抜することができた。
２．抗病性イムノバイオティクスのインビボ検証：１で選抜したイムノバイオティクスを離乳直後の子ブタに投与し、幼若期の下痢症発症度合い、成長、炎症関連指標並びにロタウイルス感染状態についてそれぞれ解析し、抗病性イムノバイオティクスのインビボ検証から、開発した評価系の有用性について検証することができた。
３．イムノバイオティクスの抗病性機構解明：PIE細胞共培養系を用いて、ロタウイルスや２本鎖RNAに対する抗病性機構の解明を進め、抗ウイルス免疫に関わる因子の遺伝子発現制御について検討することができた。
以上の研究を通して、抗病性イムノバイオティクの選抜・評価系の確立とそのインビボ検証から、本選抜・評価系の有用性を実証することができた。
