The analysis of systemic sclerosis pathogenesis with mice lacking the Fli1 gene in various cell types

Outline of Annual Research Achievements

全身性強皮症の疾病因子の一つである転写因子Fli1に着目し、loxP-Creシステムを活用して各種細胞特異的Fli1欠失マウスを作製し、強皮症の病態における各種細胞の役割について解析を進めている。
今年度は脂肪細胞特異的Fli1欠失マウス、巨核球特異的Fli1欠失マウス、gamma delta T細胞特異的Fli1欠失マウス、制御性T細胞特異的Fli1欠失マウス、骨髄細胞特異的Fli1欠失マウスに関して結果が得られた。脂肪細胞特異的Fli1欠失マウスでは、強皮症に類似した皮膚硬化と血管障害が自然発症すること、その過程にはadipocyte-to-myofibroblast transdifferentiationが関与していることが明らかとなった。巨核球特異的Fli1欠失マウスでは、モノクロタリンを投与することにより強皮症に類似した皮膚線維化が誘導できることが明らかとなった。gamma delta T細胞特異的Fli1欠失マウスと制御性T細胞特異的Fli1欠失マウスでは、野生型マウスに比してブレオマイシン誘発皮膚硬化が増強し、その過程にIL-17の産生亢進が関与していることが明らかとなった。骨髄細胞特異的Fli1欠失マウスでは、強皮症に類似した皮膚硬化と血管障害が自然発症し、さらに自己抗体産生がみられること、その過程にはM2マクロファージの活性化が関与していることが明らかとなった。
以上の検討結果より、本研究課題の目標であった8系統(血管内皮細胞、表皮細胞、B細胞、gamma delta T細胞、制御性T細胞、脂肪細胞、巨核球、骨髄細胞)の各種細胞特異的Fli1欠失マウスにおける強皮症の主要3病態の誘導について、詳細に明らかにすることができた。2019-2021年度の基盤研究Bにおいて、さらに病態解析を進めていく予定である。
