Outline of Annual Research Achievements

マラリア調査予定地であるバングラデシュは、治安当局によるイスラム過激派に対する摘発等のテロ対策が進められているものの，依然として全土にテロの脅威があり、外務省からの通達としてバングラデシュ全土レベル2（不要不急の渡航は中止）のため中止。当初の計画に従い、インドネシア島嶼群に調査地を絞り、共同研究先のインドネシアEijkman研究所（ジャカルタ）の Syafruddin博士のグループが採取したマラリア感染者の血清サンプル約3000検体を入手した。血清サンプルに含まれる抗体を検出するために、ELISA抗原6種（ハマダラカ唾液タンパクの組換え体およびヒトスジシマカのAegyptin）を大腸菌で発現・精製した。血清3000検体のELISAでの抗体価の測定の途中経過として、雨季の蚊の発生が多い時期には、抗AAPP抗体価は乾季のそれと比較して優位に上昇していた。特に、熱帯熱マラリアに感染している住民血清にはその上昇が顕著であった。興味深いことに、三日熱マラリア感染者間では低い上昇であった。これは、三日熱マラリアに特有のヒプノゾイトによる再燃感染が原因であると推察している。以上の結果から、抗AAPP抗体価を追跡調査する事によりハマダラカに対する暴露、さらにはマラリア感染アウトブレイクを予想することが可能となると期待される。追加実験項目として、ハマダラカ唾液タンパクに対する抗体価とマラリア感染の関連を動物モデルで検証することを開始した。上述の組換えタンパクおよび蚊唾液腺をマウスに免疫し、その後にネズミマラリア原虫P. berghei感染ハマダラカの吸血チャレンジ実験を行う計画で有り、現在マウスに各種タンパクを免疫中である。

バングラデシュへのマラリア調査は、治安当局によるイスラム過激派に対する摘発等のテロ対策が進められているものの，依然として全土にテロの脅威があり、外務省からの通達としてバングラデシュ全土レベル2（不要不急の渡航は中止）のため中止。当初の計画に従い、インドネシア島嶼群に調査地を絞り、インドネシア研究所との打合わせを行い、血清サンプルを入手した。血清サンプルに含まれる抗体を検出するために、ELISA抗原6種を大腸菌で発現・精製した。

Strategy for Future Research Activity

H29年度は、精製したELISA抗原6種を用いてマラリア感染率とAAPP抗体価の変動を数値化し、相関性を明らかにする。インドネシアからの留学生の協力を得て、血清サンプルを収集したインドネシア　スンバ島においてフィールド調査を実施し、マラリア感染患者からの血清サンプルの収集と住民への聞き取り調査および地図情報を記録、それぞれの家の構成人数・マラリア発症歴・保有動物種数を調査する。
