Outline of Annual Research Achievements

In this study we characterised the whole genome of representative G12P[6] strains continuously predominant in Nepal between November 2005 and January 2011. For the whole genome analysis, 52 G12P[6] strains were selected based on the electropherotypes, and the sequences were determined by the Sanger and Next Generation Sequencing methods. The whole genome sequence analysis confirmed the presence of at least four strains that corresponded to distinct electropherotypes, and their appearance was roughly in accordance with the chronology. Within the same strain the nucleotide sequence of each genome segment was highly similar, lending strong support to the classification determined by RNA polyacrylamide gel electrophoresis (electropherotyping). While the G12P[6] genotype was continuously predominant during the study period, multiple strains that were similar yet distinct in sequence at the whole genome level were noted, suggesting the continuous evolution of strains within the same G12P[6] genotype.

Next generation sequencing technique was successfully introduced in the laboratory which expedited the whole genome characterization.

Strategy for Future Research Activity

The recent collection of G12P[6] strains identified pre and post April 25, 2015 earthquake will be sequenced for their entire genome segments, and their phylogenetic relationship with the G12P[6] strains from the earlier study period will be determined. As the introduction of the rotavirus vaccine into the national immunization programme is scheduled later this year in Nepal, we will set up surveillance system for monitoring vaccine impact on the diarrhoeal disease burden due to rotavirus and any possible changes in the circulating strains.
