Elucidation of epigenetic mechanism for new therapeutic strategies against COPD

Outline of Annual Research Achievements

既にDZNepおよびEzh2コンディショナルノックアウト(cKO)マウスを用いて、エラスターゼ誘導肺気腫モデルで気腫が増悪する結果を得ている。Ezh2 cKOマウスを用いたエラスターゼ誘導肺気腫モデルにおいて、投与7日後の全肺細胞をFACSで解析したところ、NK細胞およびCD11b陽性マクロファージがEzh2 cKOマウス群でEzh2 cKO群で有意に上昇していた。同様に、DZNepを用いたエラスターゼ誘導肺気腫モデルにおいてもNK細胞およびCD11b陽性マクロファージがDZNep投与群で有意に上昇していることを確認した。また、CD11b陽性マクロファージの中でもLy6C陽性CCR2陽性細胞がDZNep投与群で増加していた。次にCCR2陽性細胞の関与を調べるために、DZNepを用いたNK細胞およびCD11b陽性マクロファージにCCR2アンタゴニストであるRS504393を用いたところ、DZNep投与による気腫増悪が相殺された。また、Ezh2 cKOマウスを用いて長期喫煙曝露モデルで検討した。喫煙開始12週間後の肺組織において、Ezh2 cKOマウス群で有意に気腫が増悪した。また、FACSではNK細胞およびCD11b陽性マクロファージが上昇していることを確認した。

Reason

エラスターゼ誘導肺気腫動物モデルおよび長期喫煙マウスモデルにおいて、効いている可能性のある細胞集団は得られたが、その後のエピジェネティクスに関与する遺伝子の特定には至らなかった。また、in vitroの系に関しては確立できておらず、ヒト検体におけるEzh2の検討を行うことができていない。

Strategy for Future Research Activity

今後は動物モデルにおけるEzh2およびヒストン修飾の検討を行うとともに、ヒト肺切除検体ならびに既存の慢性閉塞性肺疾患患者の切除肺検体および末梢血単核球細胞の遺伝子データベースを用いてEzh2の慢性閉塞性肺疾患への関与を検討する。さらに、動物モデルにおいて、Ezh2欠失後の各種免疫担当細胞の役割を検討する。
