Outline of Final Research Achievements

This study aimed to identify noninvasive biomarkers for the early detection of nonalcoholic steatohepatitis (NASH). Initially, heterogeneous nuclear ribonucleoprotein M (HNRNPM) was identified as a glycated protein from cultured cells treated with fructose for glycation. Subsequently, changes in gene expression in cells subjected to the attenuation of HNRNPM function were analyzed. The expression levels of multiple genes, including RAS guanyl-releasing protein 2, had either increased or decreased.

Academic Significance and Societal Importance of the Research Achievements

HNRNPMは、ヒトの糖尿病を模倣した培養細胞から見つかってきたものであり、NASHの原因遺伝子の候補となりえる。HNRNPMの機能の減弱によって発現が変化する遺伝子の中には、バイオマーカーとなり得るものが含まれている可能性が考えられる。これらバイオマーカーを用いてNAFLD/NASH診断用キット(簡易化したもの)を構築することができれば、大掛かりな機器や手間を必要とせず診療所レベルでの検査も期待でき、利用分野の拡大が見込まれる。
