Outline of Annual Research Achievements |
Physical exercise enhances fitness and overall health and wellness. Bouts of aerobic and anaerobic exercise induce an acute state of oxidative stress caused by an increase in reactive oxygen species (ROS), but there is little agreement which processes generate ROS and where, if they are important and how they are controlled. We produced transgenic mice expressing redox-sensitive GFP in skeletal muscle, and set up imaging systems to record muscle redox state in live anesthetized animals. Determining the conditions necessary for induction of oxidative stress during skeletal muscle activation in mice in vivo, preliminary experiments indicated that some amount of muscle fiber injury is necessary to induce oxidative stress during skeletal muscle activation. Simple electrical stimulation of muscle did not induce oxidation of the skeletal muscle glutathione pool. Injured muscle fibers became oxidized, but this oxidation did not spread to adjacent fibers in the absence of muscle activation. Experiments to publish our finding that muscle activation without injury does not result in oxidative stress were finished, but experiments using controlled injuries did not yield satisfactory results, and alternative strategies are under consideration.

Current Status of Research Progress |
Current Status of Research Progress
4: Progress in research has been delayed.
Reason
The principal investigator (PI) has prolonged his parental leave (which started from December 2018) until June 2020. As a consequence, the project is currently stopped for the duration of parental leave and will resume once parental leave ends.
|
Strategy for Future Research Activity |
The PI was on parental leave during the whole reporting period. Research will be continue once the principal investigator (PI) returns from parental leave. There is no change in research plans from last year.
研究代表者が育児休業から復職してから去年の推進方策の通り研究活動を再開します。
|
Causes of Carryover |
Delay due to parental leave. Remaining amount is planned to be used for publication costs. 育児休業によって研究活動が中断中ですので次年度使用額が生じた。
