Outline of Annual Research Achievements

Phox2b_tTA-2A-Cre Rec BAC トランスジェニックラット(①)とRosa26::floxed STOP-ChRFR(C167A)-Venus #1, #2 トランスジェニックラット(③)の掛け合わせによって生まれてくる新生児ラットから,チャネルロドプシンを発現しているものを選び実験を行った. チャネルロドプシン発現陽性新生児ラットから,脳幹-脊髄標本を作製し,第4頸髄前根(C4)から吸息性活動をモニターし,吻側腹外側延髄に青色LED光を照射し,呼吸リズムの変化を調べた.さらにこの領域の呼吸性ニューロンから細胞内電位を記録し,光照射刺激の効果を調べた.呼吸リズムは,青色LED光の照射(50ms パルス幅,150ms インターバル,5-60s )によって,3倍以上まで促進した.さらに吻側腹外側延髄に光刺激(0.5-1s)を行うことによって,呼吸リズムがリセットされた.チャネルロドプシンを発現している(つまりPhox2b陽性)Pre-Iニューロン(n=9)は,光刺激に応答して約10mVの脱分極を示した.チャネルロドプシン陰性(つまりPhox2b陰性)のPre-Iニューロン(n=7)も約5mVの脱分極を示した.一方,チャネルロドプシンを発現している吸息性ニューロン(n=5)は光刺激に応答して7.3mVの脱分極を示し,陰性の吸息性ニューロン(n=6)は有意な脱分極を示さなかった.さらにTTX存在下で,チャネルロドプシン発現ニューロン(n=8)は,光刺激に応答して約8mVの脱分極を示したが,チャネルロドプシン陰性ニューロン(n=3)は,有意な膜電位変化を示さなかった.これらの結果は,Phox2b陽性細胞から呼吸性ニューロンへの興奮性入力は,まずPre-Iニューロンにおいて統合され,その活動により吸息性バーストが周期的にトリガーされることを示唆する.

Reason

チャネルロドプシン発現ラットの新生児摘出脳幹‐脊髄標本を用いた研究では,ほぼ必要なデータがそろったと思われる.ただ,チャネルロドプシン陰性ニューロンの細胞内記録の例数を少し増やす必要がある.

Strategy for Future Research Activity

今後は,論文としてまとめるにあったって,若干不足している細胞内記録のデータを取り,解析を進める予定である.

Causes of Carryover

(理由)3月末の高松での生理学会旅費などが含まれていないため.
