Outline of Annual Research Achievements |
Fruit orchards from farms, research centers and University from whole Japan were contacted and they supplied the soil samples. In total 68 samples were collected from all Prefectures of Japan including a variety of fruits (Apple, Cherry, Grape, Kiwi, Mandarin, Chestnut, Mume Apricot, Japanese pear, Peach, Pineapple and Mango). The fruit species were chosen according to the information on Statistics of Ministry of Agriculture, Forestry and Fishery of Japan, where the top 4 producers were targeted. These soils were divided in 3 portions (Air dried, Frozen and Fridge storage). The air dried soils were submitted for chemical analysis and the freeze stored ones were used for the DNA analysis. The DNA extraction was performed in all samples, and the PCR is undergoing. Also, the enzyme analysis of the fridge stored soils in also undergoing. About the archived soils from vegetables and crops from all Japan, 180 samples had their metagenome analyzed and the bioinformatics already done. Now the comparison between the old soil chemical data is still under analysis.

2: Research has progressed on the whole more than it was originally planned.
Reason
The research is progressing according to our plans. We expected to gather the soils last year and start the analysis this year, including the metagenome and soil chemical analysis.

Strategy for Future Research Activity |
This year we expect to finish the metagenome analysis, which includes the amplicon PCR, Index PCR, Library construction and Next Generation Sequencing (Illumina MiSeq 30bp). In parallel we will carry all chemical analysis and other soil microbial activities (Enzyme analysis). Also, a software to automatize the Bioinformatics analysis is expected to be created this year.

Remarks |
This project was useful for the actual Kakenhi one, however it was fully supported by the FAPEMIG - Foundation of Support Research of the State of Minas Gerais - Brazil.
