Outline of Final Research Achievements

We define an artificial fusion protein for complexing any protein "X" to paper via the hyperthermostable chitin/cellulose binding domain (hCBD) as "X"-C Link. We have originally developed BAF-C Link using BRET-based Auto-illuminated Fluorescent protein (BAF) that reproduces the green light emission mechanism of jellyfish GFP, and developed a unique technology to combine BAF on paper.
In this study, we developed anti-GFP VHH-C Link using a specific VHH antibody against GFP in order to newly add the value of antibody function to paper. The anti-GFP VHH/paper hybrid was able to capture EGFP even after dry storage for 1 year or more at room temperature, and its application to lateral flow devices was also possible.

Academic Significance and Societal Importance of the Research Achievements

この研究は、紙という安価な構造素材に従来技術のような化学修飾手法を用いることなく、簡単に生物機能としての蛋白質機能を安定付与することで付加価値を上げる基盤技術開発である。紙にVHH抗体機能を載せることで、単なる紙素材の高付加価値化だけではなく、VHH抗体の柔軟な活用方法を新たに提示することが出来た。今回は抗GFP VHH抗体に焦点を絞ったが、他のVHH抗体でも応用可能であり、将来的には、安価な臨床検査試薬開発の基盤になり得る研究成果である。
