Clarification of causes for aberrant semen of Japanese Black beef bulls by analyses of testicular and metabolic hormones and sperm binding proteins and development of its treatment

Outline of Annual Research Achievements

平成30年度は不良精液と正常精液を持つ黒毛和種牛の血漿エストラジオール-17β、コルチゾールおよびインスリン濃度の測定して両者の比較を行った。またそれらのウシの栄養状態を比較するため、血液生化学検査を実施した。さらに12から15ヵ月齢まで1月に1回の頻度で精漿を採取して、精漿のINSL3、テストステロンおよびIGF-I濃度を測定した。加えて精子運動性の低下を示す精液にINSL3を添加して、運動性の改善がえられるか否かを検討した。
その結果、新鮮精液異常を示した雄ウシの血漿コルチゾール濃度は正常精液を持つウシに比べて14・24ヵ月齢で有意に高い値を示した。また凍結融解後精液が低受胎を示す雄ウシの血漿エストラジオールおよびインスリン濃度濃度はそれぞれ17～21ヵ月齢および11・20ヵ月齢で有意に低い値を示した。
新鮮精液異常を示す雄ウシの血清Mgは正常雄ウシに比較して6・9ヵ月齢で有意に高い値を示した。新鮮精液異常を示す雄ウシの血清AST濃度は正常雄ウシに比較して9・12・15ヵ月齢で有意に高い値を示した。高精子奇形率のみを示す雄ウシの血漿クレアチニン濃度は正常精液を持つウシに比べて9・12・15ヵ月齢で有意に高い値を示した。
雄ウシの精漿INSL3、テストステロンおよびIGF-I濃度は各月齢において新鮮精液異常と正常精液の間に有意な差はみられなかった。
精子運動性の低下を示す精液にINSL3濃度を添加した結果、運動性の顕著な改善は観察されなかった。
以上の成績から、性成熟期の黒毛和種牛の新鮮精液異常は肝酵素の血中のASTおよびストレスの指標であるコルチゾールの上昇と関連する可能性が示唆された。また凍結融解後精液が低受胎を示す雄ウシでは血中エストラジオール-17βとインスリンの低下が関与し、一方、高精子奇形率を示すウシでは血中クレアチニン増加と関連する可能性が示された。
