Outline of Annual Research Achievements

本プロジェクトでは、抗緊縮戦略を用いた新規抗がん剤の探索と、タンパク質及びメタボローム分析による作用機序の解明を目的とした。薬用植物Ferula hezarlalehzarica, Ancistrocladus abbreviatus, Calotropis gigantea, Artemisia vulgaris, Anneslea fragrans, Ancistrocladus ealaensis, Bosenbergia pandurata、Uvaria dac、Chamaecyparis obtusa、Citrus hystrix、Derris scandens, Callistemon citrinus の有効成分の抗がん作用を幅広く研究した。 本プロジェクトでは、査読付きの国際学術誌に27本の論文を発表し、国際招待講演4件、国際会議8件を含む46件の学会発表を行った。5,8-Coupled Naphthylisoquinoline Alkaloidである新規化合物Ancistrolikokine E3 は、栄養飢餓に対するがん細胞の耐性を除去する有望な抗がん剤であることが発見された。該化合物は、Akt/mTOR/オートファジーシグナル伝達経路を阻害し、膵臓がんの細胞死を誘導することが明らかになった。この発見は、ACS(米国化学協会)により重要な発見として選ばれ、世界中の数千のニュースメディアにプレスリリースされた。約130の国際的なニュースメディアがこの発見を取り上げた。本プロジェクトで発見されたnaphthylisoquinoline alkaloidsなどの化合物の多くは、がん細胞の遊走及びコロニーの形成を阻害し、抗緊縮戦略に基づく抗がん剤開発の有望な潜在的リード化合物を表している。
