新規な脂肪肝増悪因子G０S２蛋白によるインスリン抵抗性発症機序の解明

G0S2蛋白は細胞周期に関する蛋白として発見されたが、近年はATGLを抑制する作用が報告され、脂質代謝における調節因子のひとつとして注目されている。
本研究ではアデノウイルスを用いてG0S2蛋白を肝臓に過剰発現させ、G0S2蛋白が肝臓でのインスリン感受性に及ぼす影響及び作用機序について、様々なインスリン抵抗性モデル動物で検討した。
本年度は、6週齢、雌、Wistarラットを両側卵巣摘出手術もしくはSharm手術施行後、高脂肪食にて4週間飼育し、G0S2蛋白を過剰発現するアデノウイルス(Ad-G0S2)、コントロールウイルス(Ad-GFP)を投与した。投与1週間後にインスリン感受性は正常血糖高インスリンクランプ検査にて検討した。
Sharm手術群のAd-GoS2群とコントロール群においてGlucose infusion rate, Insulin stimulated-glucose disposal rate, Suppression of hepatic glucose outputに有意な差を認めず、G0S2蛋白はインスリン感受性に影響を及ぼさなかった。しかし、卵巣摘出手術群では、Ad-G0S2群は、コントロール群と比較して、Insulin stimulated-glucose disposal rateは有意な差を認めなかったが、Suppression of hepatic glucose outputが約43%減少し、肝臓でのインスリン抵抗性の増悪を認めた。
以上の結果より、肝臓でのG0S2蛋白は、肝臓でのインスリン抵抗性を増悪させるが、エストロゲンにより抑制する可能性が示唆された。

Reason
雌の高脂肪食負荷によるインスリン抵抗性状態では、肝臓のG0S2蛋白はインスリン抵抗性に影響を及ぼさなかったが、卵巣摘出によるエストロゲン欠乏状態での高脂肪食負荷によるインスリン抵抗性状態では、肝臓のG0S2蛋白は肝臓でのインスリン抵抗性を増悪させることが明らかになったことより、エストロゲンがG0S2蛋白によるインスリン抵抗性増悪作用を予防する可能性が示唆された。
