Outline of Final Research Achievements |
O-linked β-N-acetylglucosamine modification (O-GlcNAcylation) is a post-translational modification of intracellular proteins, serving as a nutrient sensor. Growing evidence has demonstrated its physiological and pathological importance in various mammalian tissues. This study examined the physiological role of O-GlcNAcylation in function and development in podocyte and proximal tubular cells. We have first confirmed that O-GlcNAcylation is necessary for maturation of podocyte foot processes, particularly after birth. In addition, we have found that protein O-GlcNAcylation is essential for renal lipolysis during prolonged fasting and offers PTECs significant protection against lipotoxicity in diabetes.

Academic Significance and Societal Importance of the Research Achievements |
本研究は糖尿病性腎症に対する新規治療標的の開発を細胞内O-GlcNAc修飾に着目し行うものである。これまで明らかとされてこなかった腎臓におけるO-GlcNAc修飾の生理的役割の解明、更には、腎症進展機構との関連性が示された。本成果は新規腎症治療標的の開発につながるのみならず、腎臓におけるO-GlcNAc修飾に関する網羅的な知見は今後の腎臓領域の研究の発展にも寄与することが期待される。
