Outline of Annual Research Achievements

前年度に引き続き、うつ病の動物モデルである成熟期ストレス（7週齢での反復拘束ストレス7日間）の（視床下部-下垂体-副腎）HPA機能について検討した。前年度までの検討で、モデル動物では血中コルチコステロン基礎値（10:00AM）が有意に増加していた。そして、脳内のグルココルチコイド受容体（GR）は変化ないが、GRの阻害因子であるFKBP5が扁桃体で有意に増加しており、この増加は抗うつ薬（SSRI）であるエスシタロプラム（ESC）10 mg/kgの2週間の反復投与によって正常レベルまで回復することを明らかにした。本年度は、遺伝子多型研究でうつ病との相関が明らかにされているCRHおよびCRH受容体（CRHR1、CRHR2）について、うつ病モデルの脳内での発現を調べた。内側前頭前野、海馬および扁桃体でCRH、CRHR1、CRHR2のmRNAをqPCRで定量したが、いずれの部位でも有意な変化は認められなかった。また、脳内FKBP5増加の臨床的な意義を明らかにするため、神戸大学精神科より提供された自殺者死後脳の前頭葉のFKBP5をウェスタンブロットで定量した。その結果、自殺者死後脳ではFKBP5が有意に低下していた（P=0.045, n=8）。この測定については現在も検体数を増やして継続中である。
