Outline of Final Research Achievements

In this research, we developed protein labeling probes for imaging protein lifetime using our original PYP-tag labeling method and a new chemical principle based on OFF-ON-OFF fluorescence switching. The regulation of intramolecular association made the probes nonfluorescent in a free state, fluorescent in a PYP-tag-bound form and nonfluorescent after protein degradation. By using one of the probes, we imaged protein expression and degradation in living cells.
