Outline of Annual Research Achievements

本研究の目的は、迅速結晶構造解析に向けた革新的な結晶化手法の確立にあり、従来の培養・精製した蛋白質の結晶化とは全く逆の発想により達成する。具体的には、(1)細胞内で鋳型となる細孔を蛋白質の結晶化により作成し、(2)構造解析の目的となる蛋白質を同じ細胞内で同時に発現させ、鋳型蛋白質結晶の細孔へ規則正しく集積し、(3)細胞に封入されたまま目的蛋白質を含む複合結晶の形成を達成した。本手法は、蛋白質結晶化反応から単離精製までを一つの細胞内で完遂する斬新な手法であり、生命原理の理解や、創薬等に向けた革新的な分子技術となる。本研究では細胞内の蛋白質結晶化反応に着目し、(1)細孔を有する鋳型蛋白質結晶の設計と細胞内合成、(2)目的蛋白質の鋳型蛋白質結晶細孔への細胞内精密集積、(3)目的蛋白質を内包した鋳型蛋白質結晶のX線結晶構造解析までを、精製した蛋白質を全く使用せず、一つの細胞内で完遂することを目指した。
本研究で達成される細胞内結晶構造解析の学術的意義は、鋳型蛋白質結晶へ集積する蛋白質に制約がなく、安定性や収率が低く結晶化が困難な蛋白質の結晶構造解析への拡張性が高い点にある。現在も様々な結晶化手法や構造解析の自動化法が開発されているものの、精製したタンパク質が必要という点では抜本的な解決策は示されていない。本研究成果は、生命現象解明の新しい分子ツールとこの長年の課題に解決策を与えるものである。
