Push / Pull non-equivalence of cells in the microchannel

Outline of Annual Research Achievements

狭窄部を含むマイクロ流路を内蔵したマイクロチップ,圧力センサ,アクチュエータ組み込み型シリンジポンプ,実時間高速ビジョン,顕微鏡,制御用PCを用いて,圧力センサフィードバック内蔵型の実験システムを構築した.特に,圧力センサ出力の介在するノイズがフィードバック系に入ると意図せぬ細胞運動を引き起こしてしまうため,ローパスフィルターを組み込み,ノイズ除去には繊細な注意を払った.この実験システムを用いて狭窄部入り口部に細胞を接触させた状態からPush/Pullの実験を行い,細胞が狭窄部に進入する際の変形の様子について実験的に調べた.Pushは入り口側の圧力を階段状に増加させることによって実現し,Pullは出口側の圧力を階段状に減らしていくことによって実現した.一般に塩分濃度が下がると細胞内部の浸透圧が上昇し,結果的に細胞の硬さが増加する.硬さの増加は細胞変形能に直結するため,Push/Pull特性にも影響を与える.この点を踏まえ,実験では液体塩分濃度を0.6%, 0.7%, 0.8%と変え,塩分濃度の影響についても考慮した.実験を行った結果,一般的な傾向として狭窄部に局所的に入り込んだ細胞の長さは,PushよりPullの方が長くなった.さらにその長さは塩分濃度が低くなるほど,短くなるという興味深い結果が得られた.塩分濃度が低くなると,細胞が硬くなる傾向があるため,変形能が低くなり狭窄部内への局所的侵入が小さくなったものと思われる.これらの実験結果は,細胞変形能評価を行う場合,Push/Pullに応じて結果が変わることを意味し,細胞変形能評価を行う場合の実験条件に十分留意する必要があることを示唆した.
