Outline of Annual Research Achievements

単独では急性骨髄性白血病(AML)を発症しないような遺伝子異常を2つ導入してはじめてAMLを発症するマウスモデルを作成し、AMLを発症させた後に片方の遺伝子の発現をサイレンシングすることによって、AMLが消失する場合としない場合とで白血病幹細胞(LSC)レベルで生じているエピジェネティックな状態を比較し、AMLの病態形成に必須のエピジェネティックな機序を同定することを試みた。
Runx1をノックアウトしたマウスでは細胞分化障害が生じ、前白血病状態を形成するが単独ではAMLを発症しない。Runx1コンディショナルノックアウトマウスの造血前駆細胞にレトロウィルスを用いてN-Ras G12D変異を強制発現させることで潜時の非常に短い劇症のAMLが発症することが報告されている。しかし発症後急速に死に至るような激烈なAMLでは今回のような経時的な解析に適さないと考えられた。そこでNRas発現量が比較的低くコントロール可能であり、また正常造血幹細胞の白血化に至る過程を明らかにするためにも、造血幹細胞への遺伝子導入可能な、テトラサイクリン誘導性レンチウィルスを用いた白血病モデルの作製を試みた。様々なMOI (multiplicity of infection)で造血幹細胞にNRasを導入することでNRas発現量を様々に調節して、レシピエントマウスに移植した。4か月の観察期間を経てもAMLの発症が見られず、レンチウィルスによる細胞毒性、発現量の不足などの要因が考えられた。現在レトロウィルスを用いたRunx1ノックアウト+誘導型NRasG12D導入白血病モデルの他、我々の研究室で確立したAMLモデルであるp53欠失JAK2V617F白血病モデルを作製したので、発症し次第がん遺伝子のサイレンシングを行うことを予定している。
