Outline of Annual Research Achievements |
脱細胞化という組織再生技術を利用してマウス子宮の再生を行った。本研究では、摘出した野生型マウス子宮に対して界面活性剤SDSによる脱細胞化処理を行い、組織から細胞を取り除いた脱細胞化子宮組織を作製し、あらかじめ子宮を部分切除する処置を行った別の野生型マウスに対して、切除部分を埋めるように脱細胞化子宮の組織片を移植する手術を行った。脱細胞化子宮組織の移植部位では1か月後に完全に子宮が再生し、移植部分の再生子宮が正常な卵巣ホルモンの組織応答能を持っているだけでなく、正常な妊娠成立・妊娠維持が可能であることが明らかになった。本技術は、幹細胞が単離されていない子宮においても、組織再生へ応用が可能な技術である点で汎用性が高い方法と考えられる。子宮再生・再構築に関与する特定の遺伝子の同定や、月経や分娩後の子宮再構築の仕組みの詳細の解明への応用が期待される。

マウスおよびヒト検体を用いた研究が順調に行われており、その成果として本年度はプロジェクトに関連した論文を複数の英文誌に発表することができた。これらの成果をさらに発展させ、次年度以降の研究の更なる進展が期待できる。
