Outline of Annual Research Achievements |
First, we completed the sequencing of the ATAC-seq libraries for each of the tissues analyzed, for two biological replicates. In total, we have generated 1,456 Million reads (PE100), amounting to a total length of 142 Gbp of clean data. Then, in order to gain a better resolution of the open chromatine regions, I have made high-quality, custom, polyclonal antibodies to perform ChIP-seq analyses against of two different factors of the Wnt pathway of the turtle Pelodiscus sinensis: LEF-1, a transcription factor (TF) active in the carapacial ridge, and β-catenin, a co-factor essential for the canonical Wnt pathway. We have performed ChIP experiments using the same tissues as in the ATAC-seq and the previous ChIP-seq for histone modifications.
